Analysis of the banding pattern of the salivary gland chromosomes of Chironomus tepperi indicates that, despite a somewhat modified male hypopygium, the relationships of this species are close to the other Australian species of the genus, particularly to Ch. oppositus. No inversion polymorphism has been found in Ch. tepperi and this, together with the relatively high chiasma frequency as measured at metaphase I, would appear to be an adaptation to provide genetic variability necessary for its colonizing ability.

The polytene chromosomes of three North American species in the subgenus Chaetolabis of the genus Chironomus are described and compared to the published karyotypes of two Palearctic species. Chironomus (Chaetolabis) atroviridis has four polytene chromosomes, like the Palearctic C. globulus, while C. ochreatus and the North American populations of C. bitumineus each have three chromosomes due to tandem fusion of arm G to arm E, as in the Palearctic populations of C. bitumineus. The results support the existence of distinct species, particularly clarifying that the names C. atroviridis, C. bitumineus and C. ochreatus cannot be synonyms, as was suggested by some previous workers. The question of whether Lobochironomus "sp. Ya6" might be conspecific with C. macani is also discussed.

The homologous sites on the salivary gland chromosomes of 13 species from three genera: Chironomus, Glyptotendipes, Kiefferulus have been mapped by means of fluorescent in situ hybridization using the evolutionary conserved gene Cpy/Cty (clone Cla1.1). In all species of genus Chironomus and genus Kiefferulus, the Cty/Cpy gene is located on arm F of chromosome EF. The relocation of the gene among the species of genus Chironomus can be done by simple or complex homozygous inversions which occurred during the divergent evolution of the chromosome of the species. In the genus Glyptotendipes, the Cty/Cpy gene was localized in arm E of chromosome EF. Since the banding patterns of salivary gland chromosomes between genus Chironomus and genus Glyptotendipes cannot be compared directly, in situ hybridization with clone of conservative gene was performed to be established some homologous chromosomes. The results obtained indicate that the chromosome arm F of Chironomus and chromosome arm E of Glyptotendipes may be homologous.

The relationships among some genera previously included as subgenera of Chironomus Meigen have been investigated using DNA sequences from two mitochondrial genes and a nuclear gene. The different mutation rates of mitochondrial genes and the nuclear 18S rDNA gene mean that they should resolve different levels of phylogeny. The analysis in large part supports many conclusions based on morphological analyses, particularly those utilizing all life stages. Einfeldia Kieffer sensu lato is confirmed as a polyphyletic grouping of species, which seems to have evolved independently, within or close to the genus Chironomus. While Einfeldia Group C, as Lobochironomus Ryser, Wülker et Scholl, is supported as a subgenus of Chironomus. Einfeldia Group B does not belong in that subgenus but rather appears to be a separate genus, for which the name Benthalia Lipina should be available. Macrogeographic patterns of polytene chromosomal banding sequences were studied in natural populations of the Holarctic species Chironomus plumosus. Of the 31 inversion sequences now known, 16 are endemic to the Palearctic, 7 are endemic to the Nearctic, and 8 are Holarctic sequences common to both zoogeographic zones. Differences in the sets of inversion sequences found on each continent, plus differing frequencies of Holarctic sequences, result in great overall divergence of karyotypes on the two continents. The karyotype of Nearctic C. plumosus differs from that of Palearctic populations primarily by the presence of a homozygous Nearctic sequence in arm A (n'plu A9), along with fixation (h'plu C2, h'plu E2, and h'plu F1), or high frequency (h'plu D2), of Holarctic sequences which are present but less frequent in the Palearctic. Identification and confirmation of Chironomus circumdatus Kieffer, 1916 (Diptera: Chironomidae) using DNA barcoding.Present study was performed to identify and confirm collected larval Chironomid up to species level. To assess molecular taxonomy and phylogeny, DNA barcoding was done using Sanger dideoxy sequencing of mitochondrial cytochrome c oxidase subunit I (COI) gene of larval sample. Bioinformatics analysis was done by using NCBI's BLAST software. By evidence of DNA barcoding, it was confirmed that present larval chironomid species was Chironomus circumdatus Kieffer, 1916.
